Does anyone have any recommendations for the best (and least congested) routes from Old City Philadelphia out to The Schuylkill River Trail? We're moving to the area and trying to figure out the best way to bike from Old City / Queen Village out to the trail that picks up towards Valley Forge. Thank you.

Yeah, Spruce is probably the most popular westbound street for cycling. It has, in theory, a "bike lane". If you squint real hard, you might be able to tell where the paint used to be. Please watch out for the potholes at Front, 2nd, 3rd, 4th, 5th,6th,7th,8th,9th,10th,11th,12th,13th,Juniper,Broad,15th,16th,17th,18th.19th,20th,21st,22nd,23rd,24th.........
Lombard would swing you around to the bottom of the South Street bridge and then hit the ramp down to the boardwalk.

Spring Garden has a bike lane too. (2 way) That'll spit you out over by the art museum. This is probably the better way as cycling on the trail south of the art museum is virtually impossible when people are out running/walking.
