I have a 2011 Cannondale Slice 5 and I can’t figure out how to adjust and tighten the rear brake.

Does the bike have a Tektro rear brake caliper? If so you can download instructions from their website: http://www.tektro.com/_english/03_support/01_download.php
It is a real pain in the ass. If you talking about the bolt that attaches it to the frame, you have to take the whole rear assembly off the frame first, tighten the bolt, then reattach it to the bike. The screw that the visible screw tightens onto is not visible when the assembly is on bike.
Mine was lose and wiggling around, and would not keep the calipers centered over the tire. I tried and cursed and drank but could not get it. I brought it to the shop and the owner played with it for a while. Once he figured it out, he called over all of his wrenches and showed them how to do it, as they had not seen this set up before and he wanted to show them how to deal with it.
Definitely not the best attachment system. I suspect that whoever designed the seat rail attachment probably had a hand in this design as well…
take the brake off the frame, tighten the adaptor that attaches to the brake, and reattach and tighten to 6nm. dont tighten the brake to the frame very hard, youll crack the carbon. if you think the brake sucks, try adjust seat angle…

Yep, definitely not the best design… it happened the same to mine when I changed the brake calipers and it took me long to set them up again.
It is tricky but possible, but if you are not very much into mechanics I’d rather take the bike to the shop so that you do not screw it up.

This is exactly what is going on with mine. I have a race on Saturday, so I guess I will have to bring it to the shop tomorrow.
There is a 3mm hex bolt that holds the rear brake mount to the frame, you can see it just above the brake. Remove that bolt an remove the brake mount, tighten the brake bolt on the underside of the mount and reattach. I find it easier to mark the position of the brake centered on the mount with a silver sharpie before removing the mount so I know where its located. I suggest using some blue locitite on the 3mm bolt when reinstalling the brake mount so it doesnt come loose.
Its essentially a copy of how the Cervelo P3C brake mounts…
