Bikes are not made of magic pixie dust. When something happens to the bike or its parts, there is a reason… take out the tube, put a couple of pumps of air into it, then submerge it in a tub of water and see where the bubbles come out. That location may have something to do with your problem. While you’ve got the tube out, run your fingers (carefully) along the interior of the tire carcass to see if there is a thorn or piece of glass stuck in the tire. Do the same for the rim to see if the rim tape has bunched up, folded, or if there is another problem. Put in a new tube or patch the old one as appropriate.
If you’ve got a tubular setup then you can do the same air bubble test, but you’d have to discard the tire if you find a leak, or else shoot the tire full of sealant and hope for the best.
