I just put the first layer of glue on the wheel and let it set for 24 hrs. I was going to apply the 2nd layer but the first layer of glue is dried and no loger tacky at all. Should I be concerned?
No - that is exactly what you want.
Well - depending on the glue… Some glues will still be tacky after 24hrs - but if you’ve used the clear Vittoria Mastik One or Conti glue then dry is just fine. If you’ve got the thick white Mastice, it will probably still be tacky. I’ve never tried the Clement red glue so I can’t tell you if it will be dry or tacky - either way should be fine. This is just your first coat that sets up the bond.

No. Not at all.

For a new tyre/clean rim I like two coats on each, both allowed to dry completely. The the last coat goes on the rim and is allowed to dry slightly (so that it’s tacky but still slightly wet) and then you mount the tyre. Partially inflate, check/adjust for straightness and then inflate to max pressure and leave overnight (24hrs is better) to “cure” or “set”.

I used the Conti glue on my new rims and tires too. I put 3 coats on the rim and 2 on the tire. Letting each layer dry completely before the next coat
