Will the Tacx Flux S smart trainer support 650 wheels? I really don’t know much about indoor trainers and if wheel size matters. Especially on this model where the rear wheel is removed.

Yes but you will need a special front wheel block.
For clarity I don't have a Premier tactical but I do have a Tacx Flux. The Flux is designed to work with 700cm road wheels and NO raiser block for the front wheel. If you move to a smaller wheel you will need to raise the front to get the height correct. This is not unique to the Flux and you will need a raiser block for 650 wheels on most smart trainers.
Its probably not relevant to you but what you can't do easily with the flux is use wheels bigger than the standard 700cm road wheel.
