I've done R2R2R and it was one of the most fun days I've ever had. Don't go in the middle of summer. Either go in the spring (April) or September/October.
We stayed at one of the Grand Canyon resort hotels, and then had a non-participating spouse drive us to the South Rim (South Kaibab Trail) at 4AM on the morning we started our run. This was the middle of October so it was 35F when we started. After 20 minutes of running (descending), we all stopped and took off our jackets because we had warmed up.
I believe it's right at 22 miles across. Almost all of this traverse is runnable, except the ascent of the North Kaibab Trail. Some people may run this but I power hiked most of it. At the North Rim, in October, there were no services available. We didn't explore very far beyond the sign, at the top, so not sure if there is a store of any kind a mile or two down the road.
Along the way, water sources are ample, you just have to make sure they are turned on. These are actual clean water spouts/spigots (or whatever you call them) and they are located so frequently along the route that you really don't need more than a 2L bladder. If they are not on, then be prepared. We took water filters with us, but never used them. There is a small stream along a lot of the trail.
On the return trip, we took the Bright Angel Trail back up to the South Rim.
Do this run. I cannot wait to do it again one day.
