Silicone in tire rim interface
This is an old school technique used to eliminate the tire rim gap for increased aerodynamics. Modern solutions being the aerowing tire and the mavic "rim ring'

My question is how do you feel this would alter rolling resistance. I would assume it might change the flexibility of the sidewall, but let's say you were careful to get it only in the gap and not up the sidewall too much?

my non scientific opinion- not much. at least not enough to bother.the flexibility of the sideall would have far more to do with the casing over which your tyre was built than any external sillicone on it. sillicone in itslef isn't a very stiff material anyway.
